Humanin is a small peptide encoded not by nuclear DNA but by the mitochondrial genome — specifically from the 16S ribosomal RNA gene. It belongs to a newly recognized class called mitochondria-derived peptides (MDPs).
Discovered in 2001 while researchers were studying genes associated with Alzheimer's disease resistance, humanin protects neurons from amyloid-beta toxicity. Its circulating levels decline dramatically with aging, which has generated significant interest in its role in age-related disease.
